l\u00f8s matrise av vannmolekyler. Hvis vann uten oksygen kommer i kontakt med en oksygenkilde, for eksempel atmosf\u00e6ren, vil oksygen sakte bevege seg inn i hullene i vannmolekylmatrisen. Oksygenet kalles oppl\u00f8st oksygen (DO), og det er det samme som \u00e5 l\u00f8se opp bordsalt i vann. Oksygen oppl\u00f8st i vann kommer fra atmosf\u00e6ren og\/eller fotosyntetiske organismer (alger) i vannet.   <\/span><\/p>\n<p>&nbsp;<\/p>\n<h2><strong>Hvorfor vil jeg m\u00e5le oppl\u00f8st oksygen?<\/strong><\/h2>\n<p><span>I likhet med mennesker m\u00e5 fisk og andre vannlevende dyr puste oksygen for \u00e5 leve, og de er tilpasset til \u00e5 puste oksygen som er oppl\u00f8st i vannet i stedet for oksygen fra atmosf\u00e6ren. Viltfisk, som \u00f8rret, krever h\u00f8ye DO-niv\u00e5er, mens grov fisk, som karpe, kan leve ved lavere DO-niv\u00e5er. Fisked\u00f8d er ofte for\u00e5rsaket av lave DO-niv\u00e5er, som ironisk nok ofte utl\u00f8ses av nedbrytning av alger som har tilf\u00f8rt oksygen til vannet f\u00f8r de d\u00f8de.  <\/span><\/p>\n<p><span>DO p\u00e5virker ogs\u00e5 den grunnleggende vannkjemien. Vann med h\u00f8y oksygentilf\u00f8rsel favoriserer den oksiderte formen av kjemikalier, mens vann med lav oksygentilf\u00f8rsel favoriserer den reduserte formen av kjemikalier. Oksygenrikt vann favoriserer for eksempel svovel i form av sulfat, SO4, mens oksygenfattig vann favoriserer svovel i form av hydrogensulfid (H2S). Sulfat er et ufarlig stoff, mens hydrogensulfid er en gift.   <\/span><\/p>\n<p><span>DO-niv\u00e5er n\u00e6r null finnes ofte i sedimentene som er akkumulert p\u00e5 bunnen av en vannforekomst. N\u00e5r oksygen krysser membranen for \u00e5 l\u00f8se seg opp i elektrolytten, forbrukes det i en kjemisk reaksjon som genererer en liten elektrisk str\u00f8m mellom de to elektrodene. Str\u00f8mmen er direkte proporsjonal med oksygenmengden i vannpr\u00f8ven. Denne metoden er n\u00e6rmere beskrevet i Standard Methods 4500-O G. Eureka tilbyr ikke lenger denne typen sensor.    <\/span><\/p>\n<p><span>Den andre typen DO-sensor er den optiske DO-sensoren, som Eurekas HDO, der et bl\u00e5tt lys rettes mot en oksygenaktiv forbindelse som er stabilisert i en oksygengjennomtrengelig polymer. Det bl\u00e5 lyset f\u00e5r den oksygenaktive forbindelsen til \u00e5 fluorescere &#8211; og   <\/span><span>Det vil si at den absorberer energi i form av bl\u00e5tt lys og deretter avgir energi i form av r\u00f8dt lys. Fluorescensen slukkes av oksygen &#8211; det vil si at utsendelsen av r\u00f8dt lys reduseres hvis det er oksygenmolekyler til stede som forstyrrer den oksygenaktive forbindelsen. Jo mer oksygen som er til stede, desto mindre r\u00f8dt lys produseres.  <\/span><\/p>\n<p><span>N\u00e5r polymeroverflaten eksponeres for vann, diffunderer oksygen inn i sensoroverflaten i henhold til oksygenmengden (&laquo;partialtrykket&raquo;) i vannet. Dermed er mengden r\u00f8dt lys som sensoren mottar, direkte relatert til oksygenmengden i vannet. Signalet fra det r\u00f8de lyset er kalibrert til de riktige DO-enhetene.  <\/span><\/p>\n<p><span>Optiske DO-sensorer har blitt standard og foretrekkes fremfor Clark-celler fordi de har liten kalibreringsdrift i felt, ikke er str\u00f8mningssensitive (ingen sirkulator er n\u00f8dvendig) og ikke krever vanskelige membranbytter som irriterer brukere av Clark-sensorer. Den 1. juli 2007 godkjente EPA ASTM International Method D888-05, Standard Test Methods for Dissolved Oxygen in Water for m\u00e5ling av DO i henhold til 40 CFR 136, noe som gj\u00f8r optiske DO-sensorer akseptable for bruk av offentlige etater. <\/span><\/p>\n<p><span>Mengden oksygen som er oppl\u00f8st i for eksempel en innsj\u00f8 eller elv, avhenger av flere variabler. Jo h\u00f8yere barometertrykk, desto mer oksygen kan l\u00f8ses i vannet. Og jo h\u00f8yere vanntemperaturen er, desto mindre oksygen kan l\u00f8ses opp i vannet.  <\/span><\/p>\n<p><span>Hvis vannet har absorbert s\u00e5 mye oksygen som det kan for en bestemt kombinasjon av temperatur og barometertrykk, sier man at vannet er mettet med oksygen. Hvis det i gjennomsnitt ikke beveger seg noe oksygen inn i vannet eller ut av vannet, sier man at oksygenet i vannet er i likevekt med oksygenet i atmosf\u00e6ren. <\/span><\/p>\n<p><span>Oppl\u00f8st oksygen (DO) rapporteres vanligvis i to enheter. DO-konsentrasjon er vekten av oksygen oppl\u00f8st i vann og rapporteres i mg\/l eller ppm. Metningsprosent er forholdet mellom oksygen i vannet og den maksimale mengden oksygen som kan oppl\u00f8ses i en vannpr\u00f8ve under de samme forholdene, og rapporteres i % metning.  <\/span><\/p>\n<p><span>De eldre Clark Cell-sensorene ble tradisjonelt kalibrert i vannmettet luft, men kalibrering i luftmettet vann blir stadig mer popul\u00e6rt. Sistnevnte gj\u00f8res ved \u00e5 riste en halv liter vann i en 1-liters beholder i ett minutt, og deretter vente ett minutt p\u00e5 at boblene stiger til overflaten og forsvinner. DO-sensoren senkes ned i vannet og f\u00e5r tid til \u00e5 stabilisere seg. Med kunnskap om vanntemperaturen og barometertrykket kan instrumentet finne ut DO-niv\u00e5et i vannet, fordi det vet at vannet er mettet med oksygen.
